This version of a blood brain barrier protein blocks many common antidepressants from entering the brain, including: amitriptyline (Elavil), citalopram (Celexa), paroxetine (Paxil), and venlafaxine (Effexor). That makes those antidepressants 7 times less effective. rs2032583 is a SNP in the ABCB1 gene (also known as the MDR1 gene), which encodes a protein that transports certain molecules across the blood-brain barrier. SNPs in ABCB1 may thus influence the intracerebral concentrations of certain drugs and thus their efficacy or potential for adverse side effects. rs2032583 is one of 9 SNPs found within a tight linkage block (r2 >= 0.8 ) such that the minor allele at any one of them predicts (with ~80%+ accuracy) that the other SNPs will also be the minor allele. The list of the 9 SNPs is shown below. When treated for depression with substrates of the protein encoded by ABCB1, carriers of one or two minor alleles at these ABCB1 SNPs have been reported to respond better than non-carriers. Moderator Emeritus manymoretodays Posted November 26, 2017 Moderator Emeritus Share Posted November 26, 2017 And a hey from me too btdt. I still wonder about Dr. Grace Jackson. Whether she is just preaching to the other doctors, which is a good thing in the long run.............and then maybe just treating the fall out from the last couple of decades. Any maybe just treating it with more medication.........albeit low doses with full disclosure. I don't know. I know she "likes" the term tardive dementia and to be honest.........I don't like her for that...... Maybe she does more though....... I sure hope so. My tardive dementia and depression comes and goes now. And I got the first longest ever stretch of stability this past year..........since I started my psychiatric patient career. So that's something. I do remember and I do spose it gets "that bad" sometimes still for me.........yet it's more doable.......my survival skills and coping somehow. I guess it's unrealistic for you to relocate to Arizona, USA. I mean it can be a great place for healing. And may be where I go, later, for retirement on a budget. I'd like to go somewhere else, if possible, for true retirement........but I am learning I don't always get to call all the shots and that's okay. You'd have to re-do citizenship and then wait for lousy medicaid benefits. I'm guessing it's better in Canada........for you........for right now anyway. I hope you can get some new glasses or whatever it takes to do a bit more time with us. In any case........keep us updated. And continue the shares as far as what you find helpful for your particular stuff now too. The MCS. Never, ever give on up on your amazing self, and healing capacities..........and faith. btdt
I still can't find 16 months of post... as stated above and I know for sure I was on her trying to find out about a drug reaction I was having in Jan 2013... can't find it not yet anyway. As for how opiates affect brain chemistry. Chemical Effects The structure and makeup of an opioid is much like that of the brain. This allows opioids to stimulate brain cell sites; prompting an increase in neurotransmitters like serotonin, dopamine, and norepinephrine https://www.h-wave.com/blog/opioids-affect-brain-mood-mental-health/ Could explain some issues I have had as after a 2012 car accident I am still taking T3 ..plus doctors could not believe the very low dose I would take and have effects. Likely they thought it was placebo ... since no doctor believes or seems to understand how a brain/body that has protracted withdrawal reacts very strangely to other medications. Apparently what I have now is multiple chemical sensitivity and that disorder too has the effect of unusual reactions to medication... oh and one more thing I learned recently ... woman don't wear bras as they find them to constricting... they actually cause me pain but that could be from old broken rib... it is always like that with me complex. Too much for doctors and too much for me. Still for some odd reason I found information years ago stating all pain meds increase serotonin and I lost the link to the info and could never find it again till today. Now that I know opiates increase all these neurotransmitters I get how opiate withdrawal can be extreme... and how people would continue to use to stop this bugger of a withdrawal. I also see how a person who has been on pain pills a long time and stops could develop conditions that require increasing neurotransmitters... it makes sense. Maybe a very low and slow taper off of opiates would allow the neurotransmitters to gradually replenish ... if the sert2 has not been damaged like ssri/snri treatment does... can't be sure as I have not read about it.. maybe I will maybe I won't... not as curious as I use to be as mostly it has not paid off... in the long run here I am so many years later. Hope this is helpful to somebody peace all WARNING THIS WILL BE LONG Had a car accident in 85 Codeine was the pain med when I was release from hosp continuous use till 89 Given PROZAC by a specialist to help with nerve pain in my leg 89-90 not sure which year Was not told a thing about it being a psych med thought it was a pain killer no info about psych side effects I went nuts had hallucinations. btdt
Nice to hear from some friends here hello ... we are all going forward even if it does not feel like it sometimes peace to you all and happy holidays I have been looking at CBD oil part of pot it can be helpful in movement disorders so I read... going to make a post here about a bit I think is important and a link to the rest if anyone is interested in reading it... 4. Beneficial Effects of Cannabinoids in the Amelioration of Nonmotor Symptoms and Progression of Parkinson's Disease 4.1. Neuroprotective Actions of Cannabinoids Cannabinoids have been shown to have neuroprotective effect due to their antioxidative, anti-inflammatory actions and their ability to suppress exitotoxicity. Plant-derived cannabinoids such as THC and CBD can provide neuroprotection against the in vivo and in vitro toxicity of 6-hydroxydopamine and this was thought to be due to their antioxidative property or modulation of glial cell function or a combination of both [182]. Studies found that CBD was able to recover 6-hydroxydopamine-induced dopamine depletion and also induced upregulation of Cu, Zn-superoxide dismutase, which is a key enzyme in endogenous defense against oxidative stress [70, 191, 227]. The reported data suggest that CBD also diminishes the increase in nicotinamide adenine dinucleotide phosphate (NADPH) oxidase expression and decreases the markers of oxidative stress, inflammation, and cell death in the kidneys [228]. Another study has also emphasized a role for superoxide anion produced by microglial NADPH oxidase in augmenting the demise of dopaminergic neurons in the PD brain [229]. The mechanism by which CBD acts to reduce NADPH oxidase expression and inhibit oxidative injury within the PD brain has yet to be confirmed but it seems to act through mechanisms independent of CB1 or CB2 receptors [76]. However, data obtained from recent studies have hinted towards a direct relationship between the CB1 receptor and mitochondrial functions in the brain [230]. The phenolic ring moieties in cannabinoids display antioxidant activity guarding against glutamate-induced neurotoxicity in a cellular model [231]. CBD produced reduction of hydroperoxide-induced oxidative damage and was more protective against glutamate neurotoxicity compared to ascorbate and a-tocopherol, indicating that CBD is a potent antioxidant [232]. Taken together, these discoveries support the hypothesis that treatment with cannabinoids having antioxidant effects may modulate mitochondrial reactive oxygen species production [233] in the PD brain. https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5165161/ Pot is legal in Canada now but the type talked about in this paper is not what your buying in the pot shops. For me the smell of pot makes me sick and contributes to my not being able to control my body... so I know something in it is toxic to me. It could be the THC part of pot that affects me or some other bit I know for sure smoking a joint would make me very sick... smelling it from person using outside starts my body jerking around. So while I am not saying go buy pot I am not saying anything really just that there may be some golden nuggets in this pot thing especially in the CBD part of the plant. Further down it talks about TRPV1 I read about here before Another receptor involved in control of movement is transient receptor potential vanilloid type 1 (TRPV1), which is expressed in sensory neurons and basal ganglia circuitry of dopaminergic neurons [126, 127]. TRPV1 receptors are molecular integrators of nociceptive stimuli activated by endovanilloids [128]. TRPV1 also interacts with ECB. In particular, anandamide is one of the major endogenous activators of TRPV1 [129–131]. Studies have revealed that motor behavior can be suppressed by the activation of vanilloid receptors [98, 99], suggesting that TRPV1 receptors might play a role in the control of motor function. For all my struggle and oversight I took an antihistamine that is a trycylic... dum dum.... seems life can always push me off a path of being drug free desperation can do many bad things to a person. Word to the wise don't waste time beating yourself up .... or a dead horse both are useless and drain much needed energy. peace all WARNING THIS WILL BE LONG Had a car accident in 85 Codeine was the pain med when I was release from hosp continuous use till 89 Given PROZAC by a specialist to help with nerve pain in my leg 89-90 not sure which year Was not told a thing about it being a psych med thought it was a pain killer no info about psych side effects I went nuts had hallucinations. btdt
For myself or anyone who has been floxed... found some nuggets in here I need to look at further https://www.hormonesmatter.com/question-doctor-lessons-learned-floxed-cipro/ August 2017: I tried taking Benfotiamine, a form of thiamine supplement, and had terrible chest pains. My chest hurt to the point that I thought I was having a heart attack. I reached out to Dr. Marrs who is a thiamine expert and asked if this was a normal reaction. Chest pains are common in people with a longstanding deficiency but due to the way they are metabolized, some forms of thiamine are tolerated better than others. Dr. Lonsdale wrote an article about the different forms of thiamine supplements, so I decided to try one that he recommended called Lipothiamine. Lipothiamine crosses the blood brain barrier, so you get the positive effects of thiamine both inside and outside the brain. Another unique attribute of Lipothiamine is that it does not require the thiamine transporter protein that enables the absorption of thiamine into cells. If you have any genetic defects in these transporters, which I highly suspect I do, then this is a great option for you. Side note – a family history of alcoholism can cause issues with thiamine transporters. Looking into your family’s health history may be helpful in putting pieces of your puzzle together. After starting Lipothiamine, my whole life changed. Not to mention my hair stopped falling out! A reoccurring thought I had been having on this journey was, “Why does life seem so hard for me when everyone else makes it seem easy?” Lipothiamine made my life easy. It was no surprise to me when I read that beriberi literally means “I can’t, I can’t.” With thiamine supplementation, I was able to live a fully functioning life for the first time in a very long time. Over the next three months, I was able to start going to a bible study, church, yoga and meditation classes, counseling and able to get a part time job on the days my children go to school. It was absolutely life changing. I did have to experiment with my dosage. I started out taking 50mg and moved up to 100mg the following week. I started having intense burning in my legs and feet, so I moved back down to 50mg. I suspect that this triggered dumping of oxalate, but that’s just a speculation based on the experience of others. Magnesium is cofactor for thiamine, so it’s important to take enough magnesium for thiamine to work properly. September 2017: After 6 weeks of taking Liptothiamine, I had some blood work done. My thyroid numbers were looking much better. I was ecstatic to see such improvement in my thyroid labs without taking medication. Things were moving in the right direction except that my absolute eosinophils were high. October 2017: I was gett January 2018: So, here we are back to present day. Overall, I’m doing really well and am trying to take things a little easier after a busy holiday season. I am not completely symptom free, but I am tremendously better than I was when this journey started. Most importantly, I am happy. I currently started supplementing with a transdermal vitamin D patch. Vitamin D is the backbone of the immune system, and I am not getting much sun lately with the cold weather. There are a few more supplements that I am interested in trying like riboflavin, biotin, vitamin A, MSM and MitoQ. In closing, I hope that my story can help spread awareness of the dangers and devastating effects of fluoroquinolines. I’m still learning new things everyday and am hoping to someday be better and healthier than I was before Cipro. I have had a funeral for my old life and am slowly finding a different and better me. I am taking this injury as an opportunity to love more deeply and hoping to help others through my experience. My best advice is to always trust in yourself, do the best you can with your nutrition, and take your thiamine. Give the rest to God. If you believe in your inner strength, you’ll see it every time you face a crisis. For more information on oxalate and the Low Oxalate Diet, please go to the Trying Low Oxalates Facebook group. Susan Owens has provided all of her research in the files section, and the moderators and members are wonderfully helpful. For more information on thiamine, please read the articles by Dr. Marrs and Dr. Lonsdale on Hormones Matter. They have also written an incredibly informative textbook called Thiamine Deficiency Disease, Dysautonomia, and High Calorie Malnutrition. Jen that was a quote from somebody else I found online... all the supplements they took. Myself I have taken a boat load of supplements and thiamine was one I started long ago for my thinking .. oddly enough I learned everything the hard way and noticed when I ate bran breakfast cereal I felt better it was so apparent I kept it in my purse... and ate it when I could not think or eat much. Then I switched to the round pellet version of all bran from the sticks and it did not work.. so I learned the sticks had a lot more thiamine.. so I bought a cheap under ten buck bottle of thiamine and took that... it worked well. Gradually over the years supplements became something I take a lot of.... had falling hair too long ago... badly biotin is suppose to be good for that.. I take it now when I have no energy take a lot of Bs individually as some mess me up I like B5 hydroxy B12 some of this is based on my 23 and me genetic report methyl folate .. ect.. Magnesium was something others here could take but not me... and what other found at paxilprogress and here is that over the course of withdrawal things change and just as they said.. things have changed and I can now take magnesium which is a god send for anxiety and muscle spasms I don't take as high a dose as others here and I have moved on to magnesium citrate but I still have magnesium malate if I think I need it... for pain more for fibro it is suggested. I take a lot of detox stuff to help multiple chemical sensitivity and avoid all perfumes in products of all types.. products without scent are not always safe as other chemicals that are unscented also cause me to have bad reactions affecting my thinking my emotions and body movements. Some I have learned from other people some I have tried based on my own research ... I mess with all the B vitamins ... get this I had a migraine coming flashing lights tried to prevent it with magnesium B2 and Q10 took it for a few days... and my hearing loss in my one ear healed up... I can now hear. Googled that and learned B2 is likely the magic bullet... so that is good. I really take a long list of things based on how I feel... emotionally mentally and physically... it all depends. \ Back to your inquiry Jen... I have looked at and never tried Benfotiamine ... it is expensive and I still don't work so have no money I take the cheap thiamine but I may try it yet... think I am doing ok on the one I take so maybe try the cheap one or eat some all bran sticks. You know you never know the lipothiamine may just do the trick for you...I copied it as food for thought and for others here. In your position I would eat some bran sticks ... if that was ok I would try the cheap thiamine ... if that was good I may actually try the other listed but would research it and talk to the doctors listed if I could. When I was in the depths of hard withdrawal most anything I tried made me worse I had to stick to food as medicine and for years that is what I did... and water therapy ... at times just water and electrolytes I don't like to think about it as it has left a huge scar I have no need to pick at as it will always be tender and a great reminder ... to just say no. For the most part I think vitamins are safe to try... a lot of them I did not do well with a some caused trouble for wks after taking only one... but I am I think the exception to most rules from what I have seen here still it is better safe than sorry you can always take more if tiny bit did not harm... you can't take away a side effect after it is in your body. \ hope that clears up some things... peace to you Jen WARNING THIS WILL BE LONG Had a car accident in 85 Codeine was the pain med when I was release from hosp continuous use till 89 Given PROZAC by a specialist to help with nerve pain in my leg 89-90 not sure which year Was not told a thing about it being a psych med thought it was a pain killer no info about psych side effects I went nuts had hallucinations. Do you still have all the anxiety, deep depression, insomnia, that people get in the early stages of wd? As it appears your stuff is physical mainly. You have done well. I have some issues with anxiety depression a big bout of that from an drug reaction ... I am keen to notice such things after what I have been thru.... life induced depression from normal things like death ect... and bad relationships and **** I have had to deal with post drug use .. reactions movement disorder ect... insomnia some.... BUT NOTHING like before.. nothing.... 4 days and nights of no sleep constant challenges of withdrawal.... no I don't have any of the above to the degree I had in withdrawal. Did it take work yes I worked my ass off trying to stay alive and keep my head above water .. I tried everything when I was able... and I have learned my body cannot tolerate these drugs... learned that from genetic testing... and I took them for 18 years .. can you say stupid loud and clear while you jump up and down and tear at the sky... it was a huge waste of my life... and a lot of regret I have given up on as it depletes me is not healing and I cannot afford the indulgence I have learned that the hard way.... I cannot afford it. I have paid enough and I am not paying one emotion more for any of this crap that was not my fault I am so over it. You can read my posts to see what helped... I can't do this much anymore... not up to it all the time. There are many many posts of mind on this site and a huge number more at paxilprogress and effexor topix that are no longer around... I have been doing this healing since 2007 that is a long time but I am still here and I am never giving up.. not ever. That is what it takes to get thru this ... as an old wise women once said when your going thru hell just keep going. wishing you peace Leo WARNING THIS WILL BE LONG Had a car accident in 85 Codeine was the pain med when I was release from hosp continuous use till 89 Given PROZAC by a specialist to help with nerve pain in my leg 89-90 not sure which year Was not told a thing about it being a psych med thought it was a pain killer no info about psych side effects I went nuts had hallucinations. As I had a head injury and was diagnosed with a concussion in 85 I was sent to a head injury clinic in 1990 five years after the accident. I don't think they knew I had been on prozac I did not think it a big deal and never did finish the bottle of pills. I had tests of course lots of them. Was put into a pain clinic and given amitriptyline which stopped the withdrawal but had many side effects. But I could sleep something I had not done in a very long time the pain lessened. My mother got cancer in 94 they switched my meds to Zoloft to help deal with this pressure as I was her main care giver she died in 96. I stopped zoloft in 96 had withdrawal was put on paxil went nutty quit it ct put on resperidol quit it ct had withdrawal was put on Effexor... 2years later celexa was added 20mg then increased to 40mg huge personality change went wild. Did too fast taper off Celexa 05 as I felt unwell for a long time prior... quit Effexor 150mg ct 07 found ****** 8 months into withdrawal learned some things was banned from there in 08 have kept learning since. there is really not enough room here to put my history but I have a lot of opinions about a lot of things especially any of the drugs mentioned above. btdt
Jen because you mentioned it I have looked further into the site about thiamine thank you. This may be my missing link... we all help each other all the time.. never fails to amaze me... I had already forgotten about the hormones matter page... please take a look at this page I found today... you just never know where you are going to find healing like it is the charm on survivor... ha... under a rock.. a log .... I know it not called charm but I cannot think of the word it is called.. just keep turning over rocks and be careful what people want you to pay for cause I think and entire industry is supported by people like us maybe several industries.... and I am tried of being a slave to them and giving them all I ever had... very tired of that... then again whatever gets you thru the night some days... isn't that what got us here to start with... k now I am doing my own brand of bs so I will shut up... do what you need to and so will I. https://www.hormonesmatter.com/ttfd-thiamine-derivative/ that link is more for me than anyone else... peace all WARNING THIS WILL BE LONG Had a car accident in 85 Codeine was the pain med when I was release from hosp continuous use till 89 Given PROZAC by a specialist to help with nerve pain in my leg 89-90 not sure which year Was not told a thing about it being a psych med thought it was a pain killer no info about psych side effects I went nuts had hallucinations.
